Anti-LIN-9 [mAb#6] Antibody

This mouse IgG1 monoclonal antibody was generated against a purified recombinant LIN-9 short-GST fusion protein and reacts with human and mouse LIN-9 (epitope: aa 360-542).

LIN-9, also known as TGS, BARA or TGS1, is a 542 amino acid protein that localizes to the nucleoplasm and is a mammalian homolog of the C. elegans Lin-9 protein. Expressed in testis and thymus, LIN-9 functions as a component of the DREAM complex (also known as the LINC complex), which is comprised of several proteins, all of which work in concert to repress cell cycle-dependent genes. LIN-9 specifically acts as a tumor suppressor that associates with Rb and inhibits DNA synthesis, possibly also controlling the expression of genes that are required for the G1/S cell cycle transition. Three isoforms of LIN-9 exist due to alternative splicing events.

Characterization of LIN-9 mAbs

(A)Two independent mAbs were produced by immunizing BALB/c mice with a full-length GST-BARA/LIN9-L. Cell lysates obtained from 293T cells were immunoprecipitated with control IgG1 (CT), mAb#1, mAb#6 or an anti-BARA/LIN-9 pAb. Western blotting was sequentially performed with anti-BARA/LIN-9 mAb #6, mAb#1 and pAb (top, medium and bottom panels, respectively). The heavy chain of the IgG (IgG-HC) is indicated by an asterisk. The IgG-HC for mAb#1 migrates as a doublet as demonstrated by direct Western blotting in the absence of lysates (data not shown). The anti-BARA/LIN-9 pAb and mAb#6 immunoprecipitate proteins with a molecular mass of 62 and 55 kDa (long and short forms, respectively) that are detected by blotting with mAb#6 and pAb (upper and bottom panels, respectively). The mAb#1 can only recognize BARA/LIN9-L.(B) Expression of BARA in cell lines. Cell lysates (1 mg) from the indicated cell lines were immunoprecipitated with a rabbit anti-BARA/LIN-9 antibody (BARA) or normal rabbit serum (NR), followed by SDS-PAGE (8% gel), and Western blotting with the anti-BARA/LIN-9 monoclonal antibody (mAb) #6. The migration of the long and short forms is indicated by an asterisk. IgG, immunoglobulin heavy chain. The 68 and 50 kDa molecular weight markers are shown.
