Diazo Biotin-Alkyne

An azide-reactive, sodium dithionite (Na2S2O4) cleavable biotinylation reagent.


  • Cleavable – a diazo spacer arm allows protein targets to be released under mild conditions
  • Chemoselective – azide and alkyne groups do not react or interfere with other functional groups found in biological samples but conjugate to one another with high efficiency
  • Solubility – reagent easily dissolves in water-miscible solvents for subsequent dilution into aqueous reaction mixtures
  • Extended PEG3 spacer – reduces aggregation, minimizes steric hindrance for affinity-binding to avidin or streptavidin

Diazo Biotin-Alkyne allows labeling of azide-tagged biomolecules via a copper-catalyzed click reaction. Biotinylated protein targets are selectively captured through conventional affinity purification protocols that exploit the biotin - streptavidin interaction. Captured protein targets can be release of under mild conditions with 50 mM sodium dithionite solution, and identified by mass spectrometry, ELISA, dot blot, or Western blot applications.
