Cy3 - Methyltetrazine

Cy3-Tetrazine is a fluorescent probe useful for chemoselective labeling and detection of TCO-tagged molecules.

Highlights:

  • Biocompatible – click reaction occurs efficiently under mild buffer conditions; requires no accessory reagents such as a copper catalyst or reducing agents (e.g. DTT)
  • Chemoselective – tetzines and trans-cyclooctene groups do not react or interfere with other functional groups found in biological samples but conjugate to one another with high efficiency
  • Unprecedented kinetics – inverse-electron demand Diels-Alder chemistry is the fastest bioorthogonal ligation available, delivering conjugates within 30 min at 1 mg/mL concentrations

Tetrazine-activated fluorescent dyes are well suited for the detection of TCO-labeled biopolymers. The inverse-electron demand Diels-Alder cycloaddition reaction of TCO with tetrazines is a bioorthogonal reaction that possesses exceptional kinetics (k > 800 M-1s-1) and selectivity. Such excellent reaction rate constants are unparalleled by any other bioorthogonal reaction pair described to date.
