The copper-catalyzed azide-alkyne cycloaddition (CuAAC) has been widely utilized in applications of bioconjugation. The CuAAC reaction involves the coupling of an azide and a terminal alkyne to form a stable triazole linkage between two disparate molecular entities. In addition to the CuAAC reaction, the catalyst free strain-promoted azide-alkyne cycloaddition (SPAAC) is emerging as a dominant method for bioconjugation. Unlike the CuAAC reaction where a terminal alkyne is requisite, SPAAC makes use of an internal cyclic alkyne with sufficient ring-strain to promote the cycloaddition with its cognate azide. While these coupling processes are straight forward neither azides nor alkynes occur naturally in biomolecules.
