Mouse Fibroblast, COX10 Knock-Out Cell Line

Mouse fibroblast cell line lacking the gene cytochrome c oxidase assembly factor heme (COX10).

Cytochrome c oxidase (COX), the terminal component of the mitochondrial respiratory chain, catalyzes the electron transfer from reduced cytochrome c to oxygen. To generate the cell line, a primary skin fibroblast culture was stablished from a knockin mouse homozygous for the floxed COX10 (exon 6). Cells were immortalized by retroviral infection of E6-E7 type 16 papiloma virus carrying G418 resistance. To generate COX10 KO, immortalized cells were transfected with plasmid expressing cre recombinase and clones were obtained after hygromycine selection. This is KO clone KO#19. Generation and characterization of cell line is described in PMID: 16782876. Mouse used to generate cell line had mixed genetic background (129svj and C57BL6J).
