poly(GMA-ran-OEGMA) Enzyme Stabilization Reagents

Enzyme-polymer conjugation tools, based on the cylindrical molecular brush architecture, for increased enzyme stability.

The copolymer (PL) has ~34mol% of epoxide side groups (glycidyl methacrylate, GMA) and ~66mol% of Poly (oligo(ethylene glycol) methacrylate (POEGMA, Mn 950) side groups, abbreviated as poly(GMA-ran-OEGMA). The copolymer is capable to react with multiple amino groups (of lysine, Lys) present in a typical enzyme structure creating an enzyme-polymer conjugate via 'grafting to' approach. PL is water soluble and has molecular weight to allow for enveloping of multiple enzymes by single PL macromolecule that enhances the enzyme activity by 2 to 5 fold and, in some cases, even thermal stability up to 100C. The thermal stability of enzymes is tested with lysozyme (up to 150C) and trypsin (up to 100C). The copolymer is of great interest to improve the industrial enzymes.
