Anti-Citrullinated Human Fibrinogen [10E9.3] Antibody

This mouse monoclonal antibody was generated against citrullinated-human fibrinogen and recognizes citrullinated-human fibrinogen.

Fibrinogen is a plasma clotting factor composed of three different polypeptide chains (α, β and γ). Under chronic inflammatory conditions, fibrinogen can be acted upon by the enzyme peptidylarginine deiminase 4 (PAD4), converting specific arginine residues to citrulline. Patients with rheumatoid arthritis (RA) produce antibodies that bind citrullinated human fibrinogen. The citrullinated fibrinogen and reactive antibodies form immune complexes in RA patients, and these immune complexes can stimulate macrophages. Recently, it has been discovered that immunizing certain strains of mice with citrullinated human fibrinogen can induce the production of anti-citrullinated human fibrinogen antibodies and the development of arthritis that shares many pathophysiological features of the human disease.

Research
Balb/cJ mice were immunized i.p. with 100 ugs of citrullinated-human fibrinogen glutaraldehyde-coupled to KLH, and emulsified 1:1 in Complete Freunds Adjuvant. 22 weeks later, mouse was boosted i.v. with 100 ugs of citrullinated human fibrinogen coupled to KLH in PBS. Four days later the spleen was harvested and spleen cells fused to NS0 myeloma cells. Hybrids were screened by an ELISA assay on citrunillated human fibrinogen or unmodified human fibrinogen coated plates and developed with HRP-Goat-mouse IgG. Hybridomas from positive wells were cloned by limiting dilution.
