Anti-LINE-1 Retrotransposable Element ORF2 Protein [1G4E11] Antibody

This mouse IgG1k monoclonal antibody was generated against bacterially-purified human endonuclease of open reading frames 2 (ORF2) and is specific for human Long Interspersed Nuclear Element-1 (LINE-1) ORF2.

Highlights:

  • Recognizes human LINE-1 ORF2
  • Suitable for Western Blot applications

LINE-1 retrotransposable element ORF2 protein (ORF2p, LINE1 (also L1 and LINE-1) has a reverse transcriptase activity required for target-primed reverse transcription of the LINE-1 element mRNA, a crucial step in LINE-1 retrotransposition. LINE1 also posesses endonuclease activity, allowing for the introduction of nicks in the chromosomal target DNA. The protein cleaves DNA in AT-rich regions between a 5' stretch of purines and a 3' stretch of pyrimidines, corresponding to sites of LINE-1 integration in the genome.

Comments
Western Blot Protocol: We recommend using gels that have a great separation of the 75-200 kDa range for proteins. The 4% Tris Glycine gels from Invitrogen (EC6055BOX), the Bio-Rad 5% Tris HCl gel (3450001) or the Bio-Rad 7.5% Tris HCl (3450005) all work well in separating non-specific bands caused by the secondary antibodies. We had tried several secondary anti-Mouse antibodies and all of them had a non-specific band near the size of ORF2p. If you have an anti-Mouse secondary that does not cause a non-specific band(s) around 120-150kDa then your choices are greatly expanded. Full protocols can be found in our publication: Sokolowski M, et al. Mob DNA. 2014 Dec 10;5(1):29. See ‘For SDS Tris Glycine gels’ under the western blot analysis heading of the material and methods section. Adding 4mL of NIH-3T3 media (cultured for 24 hours) and 11mL of 3% blotto in the blocking buffer and the secondary antibody greatly reduces non-specific bands in HeLa and NIH-3T3 cells.
