STAT2-Defective Mutant Daudi Cells (RST2)

Interferon (IFN) therapy for certain cancer types has proven successful, however resistance often develops. The RST2 cell line was developed to study IFN resistance. This line was isolated from the highly IFN-sensitive Daudi cell line by growth in the continuous presence of IFN.

RST2 cell line characteristics:

  • Good for difficult to propagate  viruses
  • Resistant to antiviral, antiproliferative, and gene-induction actions of IFNa
  • Deficient STAT2 protein expression due to alternative splicing within the intron between exon 19 and 20
  • Shows intrinsic resistance to apoptosis induced by chemotherapeutic agents (camptothecin, staurosporine, and doxorubicin)
  • Expression of STAT2 rescues cell sensitivity to IFNs and chemotherapeutic agents

IFN induced antiviral activities in Daudi and RST2 cells1:

Daudi and RST2 cells were treated overnight with the indicated concentrations of IFNa. The cells were then infected with VSV at 0.1 plaque-forming unit/cell, and the virus yield in the medium was determined by plaque formation.
