4PD in vivo MDSC/Mφ Transfection Kit

4PD in vivo transfection kit is a nanocarrier for the selective drug delivery to and transfection of myeloid-derived suppressor cells (MDSCs) and macrophages (Mφ) in vivo.

Highlights:

  • 4PD is a G5 PAMAM dendrimer conjugated to a peptide specifically targeted to MDSCs and tumor associated macrophages
  • The kit allows for transfection of primary human MDSC and macrophages
  • Nucleic acids can be delivered in vivo, eliminating the need to isolate and enrich MDSCs
  • The kit allows the simultaneous in vivo delivery of multiple nucleic acids to MDSCs and macrophage
  • Simple and quick procedure; nucleic acids or drugs and dendrimer selfassemble into active complex in five minutes
  • The low immunogenicity of the optimize formulation allows multiple administration without the induction of neutralizing antibody
  • The use of 4PD in conjunction with BrUTP allows to follow the fate of transfected cells by flow cytometry.
  • Quality Controlled - Each lot is tested in vivo for its ability to preferentially transfect myeloid cells using AF555-control shRNA and ex vivo flow cytometry analysis

Myeloid-Derived Suppressor Cells (MDSCs) and tumor-associated macrophages (Mφ) are emerging as the most important pro-tumoral populations present in tumor host. These cells play a key role in promoting tumor progression and metastasis as well as in inhibiting spontaneous or therapeutically induced anti-tumor immunity. The 4PD nanoparticle is a G5 PAMAM dendrimer functionalized with a peptides recognizing receptors on MDCSs and macrophages. The formulation has been optimized with the use of amphipathic molecules that increase specificity and reduce the immunogenicity of the nanoparticle. Nucleic acids (e.g., DNA, RNA, shRNA), small molecules, drugs, etc. are loaded onto the dendrimer and the amphipathic molecule is added to reduce immunogenicity. 4PD has been shown effective in specifically targeting and delivering nucleic acids to MDSCs and serves as a useful tool for detection or the modulation of gene expression.
