Anti-Complement Factor H, 402Y [1H10] Antibody

This mouse IgG1k monoclonal antibody was generated against a synthetic peptide corresponding to human complement factor H, polymorphic variant 402-Tyr and is specific for human complement factor H (FH-402-Tyr).

The complement system consists of a number of small proteins found in the blood, generally synthesized by the liver, and normally circulating as inactive precursors (pro-proteins). When stimulated by one of several triggers, proteases in the system cleave specific proteins to release cytokines and initiate an amplifying cascade of further cleavages. The end-result of this activation cascade is massive amplification of the response and activation of the cell-killing membrane attack complex.

Variants of complement pathway associated genes are significantly associated with age-related macular degeneration (AMD), the leading cause of blindness in the elderly. Most notably, the Y402H variant of human complement Factor H (FH) was identified as a common susceptibility allele and may be responsible for up to 50% of all AMD cases.

0.4 ul plasma or 250 ng purified Factor H from blood donors homozygous for either FH-402H (FH-H) or FH-402Y (FH-Y) were separated by SDS-PAGE and probed by immunoblotting with anti-FH-H mAb 23F8, anti-FH-Y mAb 1H10 or anti-FH mAb C18/3 which reacts with both FH variants equally well.
