Anti‐IQGAP2 [A2] Antibody

This IgG2a mouse monoclonal antibody was generated against purified, recombinant, N-terminally his-tagged human IQGAP2 (isoform 1), and is reactive towards human and mouse IQGAP2.

IQ motif containing GTPase activating protein (IQGAP) is a scaffolding protein that interacts with cytoskeleton, cell adhesion molecules, and with signaling molecules to regulate cell morphology and motility. The protein contains three IQ domains, one calponin homology domain, one Ras-GAP domain, and one WW domain. It has been known that IQGAP2 binds to activated CDC42 and RAC1 but does not seem to stimulate their GTPase activity. IQGAP2 is found in the liver, stomach, and platelets and is over 50% identical to IQGAP1, but appears to have a drastically divergent function in terms of pathology.

Immunofluorescence

CHO-K1 (Chinese hamster ovary) cells transfected to express the indicated fluorescently tagged fusion proteins derived from human IQGAP2 or IQGAP1 were stained by immunofluorescence with mouse monoclonal anti-IQGAP2 (clone A2) followed by AlexaFluor 594-goat anti-mouse IgG. Note that anti-IQGAP2 labeled the N-terminal 1/3 (amino acids 1-528) and full length (FL) versions of IQGAP2, but did not label FL IQGAP1.

Western Blot

An unfractionated homogenate of AG5 human gastric carcinoma cells was labeled with anti-IQGAP2 (clone A2). A single band in the MW range of full length IQGAP2 (~174,000) was labeled.

Protein Expression in Cells
