Leptin-Related Synthetic Peptides

These small synthetic peptides encompass an active epitope of leptin (amino acid residues 116-122) and can mimic the effects of leptin on energy balance and glucose regulation.

Leptin, the encoded protein from the ob gene, is a secreted hormone produced principally by white adipocytes (fat cells). Leptin reduces food intake and body weight gain, normalizes glucose and insulin levels, promotes fat metabolism, stimulates energy expenditure and thermogenesis, and increases insulin sensitivity in leptin-deficient ob/ob mice.

Protocol

It is recommended that the peptide be reconstituted immediately before use. The peptide solution cannot be stored at room temperature or refrigerated for more than 24 hours without losing activity.

OB3: Leptin-like activity when delivered IP in PBS to genetically obese ob/ob and db/db mice at a minimal effective concentration of 40 mg/kg, once daily.

[D-Leu-4]-OB3: Leptin-like activity when delivered orally in dodecyl maltoside to genetically obese ob/ob and db/db mice at a minimal effective concentration of 20 mg/kg, twice daily.

Comparison of OB3 and [D-Leu-4}-OB3

Effects of seven daily injections (1 mg/day, ip) of OB3 and its D-amino acid-substituted analog [D-Leu-4]-OB3 on body weight gain in female C57BL/6J ob/ob mice. The graph shows the changes in body weight (expressed as a percentage of initial weight) in mice treated with vehicle, OB3, or [D-Leu-4]-OB3. Each value represents the mean ± SEM change in body weight for a group of 6 mice.
