Anti-NOX2/gp91phox [54.1] Antibody

This IgG1 mouse mononoclonal antibody was generated against the partially purified human neutrophil flavocytochrome b (cytochrome b559 or cytochrome b558) and is reactive against human gp91phox amino acids 382-389.

Gp91phox (portion of the heterodimer Cytochrome b) is a critical component of the membrane-bound oxidase of phagocytes that generates superoxide. It is the terminal component of a respiratory chain that transfers single electrons from cytoplasmic NADPH across the plasma membrane to molecular oxygen on the exterior. It also functions as a voltage-gated proton channel that mediates the H(+) currents of resting phagocytes. Defects in the CYBB gene are a cause of X-linked chronic granulomatous disease (X-CGD). X-CGD is characterized by the failure of activated phagocytes to generate superoxide and patients suffer from life-threatening bacterial/fungal infections.

Specificity of mAb for cytochrome b and phage pIII proteins. Intact phage expressing known amino acid sequences (lanes B, C, E, and F) and partially purified cytochrome b (lanes A and D) were separated on SDS-PAGE polyacrylamide gels and transferred to nitrocellulose. Samples were Western blotted with mAb 54.1 (lanes A-C) or mAb 44.1 (lanes D-F). Lanes B and F contain proteins of phage sequence 3 (PQVRPI), and lanes C and E contain proteins of phage sequence 27 (FKRGVD.
