Luciferase-Expressing B Lymphocyte Cell Line (A20)

A20-Nluc-copGFP (A20) is a mouse B cell lymphoma cell line expressing nano-luciferase (Nluc) established from spontaneous reticulum cell neoplasm.

Highlights:

  • Useful for in vivo imaging via stable expression of Nluc
  • Nluc is an ATP independent luciferase, 150X brighter, more stable, and smaller in size compared to renilla or firefly luciferase

To prepare, A20 cells were transduced with lentiviral particles containing plasmid pCDH-CMV-Nluc-P2A-copGFP-T2A-Puro which encodes Nluc, copGFP and puromycin. Stable Nluc expressing polyclonal population was generated by puromycin selection and further confirmed by copGFP expression. Nluc expression was confirmed by measuring luminescence using substrate Vivazine. In vivo engraftment of these cells are confirmed in NSG mouse by tail vein injection followed by bioluminescence imaging.
