DBCO-Acid

An amine-reactive building block used to derivatize primary amines with DBCO.

Highlights:

  • Biocompatible – click reaction occurs efficiently under mild conditions; requires no accessory reagents such as a copper catalyst or reducing agents (e.g. DTT)
  • Chemoselective – azide and DBCO groups do not react or interfere with other functional groups found in biological samples, but conjugate to one another with high efficiency
  • Primary amine reactivity – modifies amine-containing molecules with DBCO in the presence of activators
  • Short spacer – 4 carbon atom spacer arm keeps DBCO moiety close to tagged molecules

DBCO reagents, also known as ADIBO or DIBAC are routinely used building blocks for synthesis of complex molecules to be used in Cu-free ligation applications. Using DBCO-Acid primary or secondary amines can be easily functionalized with DBCO moiety in the presence of activators (e.g. EDC, or HATU) through a stable amide bond. Short spacer arm adds minimal mass to modified molecules (305.11 daltons).
