Rac1 Antagonist (W56)

W56 comprises residues 45-60 of the ß3 region of Rac1. W56 targets the Trp56 site of Rac1, and competes for GEF binding. W56 (concentrations >200 µM) selectively inhibits Rac1 interaction with Rac1-specific GEFs: TrioN, GEF-H1, and Tiam1.

Rac1 is a member of the Rho subfamily of the Ras small GTP-binding proteins. Rac1 has been studied for its role in actin cytoskeleton reorganization, transcriptional activation of genes, stimulation of DNA synthesis, endocytic and exocytic membrane trafficking, regulation of translation, and pathologies such as cancer. Rac1 is tightly regulated by GTP binding, which is facilitated by guanine nucleotide exchange factors (GEFs). Blocking Rac1/GEF binding effectively inhibits Rac1 activity.
