Rat Cochlear Microglial Cell Line (Mocha)

Mocha cells are a rat cochlear microglial cell line. They were developed from an immunomagnetically enriched population of CD11b+ cells from P3 rat cochlea and immortalized with the 12S E1A gene of adenovirus in a replication-incompetent retroviral vector.

Highlights

  • Express markers consistent with microglia (CD68, CD11b, Iba1, F4/80, and TMEM119)
  • Respond to LPS stimulation by upregulation of genes (Cox2, ICAM-1, Il6r, Ccl2, Il13Ra, and Il15Ra) as well as release of cytokines (IL-1beta, IL-12, IL-13 and RANTES)
  • Phagocytose fluorescent beads at 37C, a hallmark of microglial function
  • Have undergone verification by IDEXX-RADIL to be of rat origin without contamination by other mammalian cell types. A baseline genetic profile of these cells is available here: Seigel, et al. Mol. Cell. Neurosci. 85 (2017) 202-210.

Microglia are a unique lineage of mononuclear phagocytic cells that play an essential role in development, immunological defense and in- flammatory processes in the central nervous system. Microglia account for 10-15% of all cells found within the brain. As the resident macrophage cells, they act as the first and main form of active immune defence in the central nervous system (CNS).
