NuLink Alkyne

NuLink Alkyne copper-catalyzed azide-alkyne cycloaddition (CuAAC) reagent.

Highlights:

  • Rapid - Alkyne incorporation can be complete in less than 15 min with clean product ready in under 1 hr
  • Quantifiable - Alkyne incorporation is quantified by a simple single-point U.V. scan (335 nm)
  • "Click" Monitoring - By using NuLink Alkyne with one of NuChemie's quantifiable azides, UV monitoring can be used to track reaction progress of the azide-alkyne cycloaddition
  • Dual Monitoring - NuLink Alkyne renders labeled molecules with unique absorbance (335 nm) and fluorescent (420 nm) signatures
  • Minimal byproducts - NuLink regents produce only inert CO2 upon reaction. Unreacted or hydrolyzed reagent is easily removed by means of a number of downstream processing steps
  • Water Soluble - Highly water soluble (≥ 40 mg/ml)

The copper-catalyzed azide-alkyne cycloaddition (CuAAC) has been widely utilized in applications of bioconjugation. The CuAAC reaction involves the coupling of an azide and a terminal alkyne to form a stable triazole linkage between two disparate molecular entities. In addition to the CuAAC reaction, the catalyst free strain-promoted azide-alkyne cycloaddition (SPAAC) is emerging as a dominant method for bioconjugation. Unlike the CuAAC reaction where a terminal alkyne is requisite, SPAAC makes use of an internal cyclic alkyne with sufficient ring-strain to promote the cycloaddition with its cognate azide. While these coupling processes are straight forward neither azides nor alkynes occur naturally in biomolecules. NuChemie’s azide-alkyne reagent set is the simplest easy-to-use method for the incorporation and quantification of both azides and alkynes into almost any biomolecule platform.
