Anti-Streptavidin [3A20.2] Antibody

This mouse monoclonal antibody was generated against and recognize streptavidin.

Highlights:

  • Reacts with streptavidin
  • Suitable for ELISA applications

Streptavidin is widely used in biotechnology and molecular research for detection, purification, crosslinking, and labeling of biotinylated targets. However, wild-type streptavidin is an obligate tetramer and can crosslink biotinylated targets. Target aggregation is a significant obstacle in some applications, including the labeling of biotinylated cell surface receptors, where crosslinking can perturb protein stability and function.

Research
Balb/c mice were immunized i.p. with 100 ugs of Streptavidin, emulsified 1:1 with Complete Freunds Adjuvant. After 7.5 weeks, mouse was boosted i.v. with 100 ugs of streptavidin in PBS. Four days later the spleen was harvested and spleen cells fused to NS0 myeloma cells. Hybrids were screened by an ELISA assay on plates coated with SA bound to biotinylated ovalbumin, and developed with HRP conjugated goat anti mouse Ig. Hybridomas from positive wells were cloned by limiting dilution.
