Anti-Clostridium Perfringens Epsilon Toxin [4D6] Antibody

This mouse IgG monoclonal antibody was generated against epsilon Toxoid (A single amino acid mutant of whole toxin) and recognizes clostridium perfringens epsilon toxin.

Highlights:

  • Reacts with C. perfringens epsilon toxin
  • Detects multimer forms of protein
  • Able to detect as little as 625pgs of recombinant N-terminal His-tagged epsilon mutant toxoid
  • Detects native un-cleaved ETX proto-toxin as well trypsin cleaved fragments that have higher electrophoretic mobility
  • Useful as a diagnostic tool to detect Epsilon toxin a classified bio-terrorism agent
  • Suitable for Western Blot applications

The epsilon toxin is one of 12 protein toxins produced by Clostridium perfringens, a Gram positive, anaerobic spore-forming rod. There are five strains of C. perfringens, designated A through E. Each strain produces a unique spectrum oftoxins. The epsilon toxin is made by types B and D. This toxin is a pore-forming protein; it causes potassium and fluid leakage from cells. In addition to the epsilon toxin, C. perfringens type D strains produce alpha toxin and type B strains produce alpha and beta toxins. C. perfringens type B causes severe enteritis in young calves, foals, lambs and piglets. Type D causes enterotoxemia in sheep and goats and, on rare occasions, in cattle. All five strains can infect wounds in any species.

Western Blot

Purified 4D6 anti-ETX antibody is used in this western blotting at a final concentration of 0.5ug/ml. They used Anti-Mouse-IgG-HRP as secondary antibody at 1:3000 dilution. Western is developed using TMB, liquid substrate system for membranes. Lanes 1: 625pg of His-Epsilon Toxoid, Lane 2: 1.25ng of His-Epsilon Toxoid, Lane 3: 2.5ng of His-Epsilon Toxoid, Lane 4: 5ng of His-Epsilon Toxoid, Lane 5 : 10ngs of His-Epsilon Toxoid, Lane 6: SeeBluePlus2 Protein-Marker, Lane 7: Semi-pure, ~ 5ngs ETX proto-toxin, Lane 8: Trypsin activated, 5ngs of ETX proto-toxin, Lane 9 : Trypsin activated, 2.5ngs of ETX proto-toxin.
