Anti-JARID1B [4440] Antibody

This polyclonal antibody was generated against a GST-JARID1B polypeptide and is specific for human JARID1B.

JARID1 (Jumonji AT-rich interactive domain 1) family members include two highly related genes JARID1A/RBP2/KDM5A and JARID1B/PLU1/KDM5B, and two less related genes JARID1C/SMCX/KDM5C and JARID1D/SMCY/KDM5D, on the X- and Y-chromosomes, respectively. The four JARID1 proteins and their orthologs in other organisms have been described as histone demethylases specific to methylated lysine 4 of histone H3 (H3K4). In the absence of JARID1A, JARID1B may be recruited to some of JARID1A target regions. High expression of JARID1B mRNA and protein has been found in all breast cancer cell lines examined, in cells cultured from primary breast cancers and in the invasive and in situ components in primary breast cancers. In breast cancer cells, JARID1B exhibits oncogenic properties and has been linked to repression of BRCA1 and other tumor suppressor genes.

Immunoprecipitation (IP) and IP-Western experiments

JARID1B

A. Performance of JARID1B rabbit anti-sera in immunoprecipitation experiments. SAOS-2 osteosarcoma cells were transfected with a HA-PLU1 overexpression plasmid and immunoprecipitation reactions were performed with antibodies 4439 and 4440 (at 1:1,000 dilution) or the corresponding pre-immune sera overnight at 4°C. Positive control included anti-HA hybridoma 12CA5. Negative controls included unrelevant antibodies 4437 and 4439, their pre-immune antiserum, rabbit IgG, as well as immunoprecipitations performed with lysates from cells transfected with the vector. The immunoprecipitates were separated by 6.25% SDS-PAGE and immunoblotted with the HA antibody 12CA5. The JARID1B -specific band is just below 180 kDa. B. Performance of JARID1B rabbit anti-sera in IP-Western experiments. SAOS-2 osteosarcoma cell lysates were incubated with the indicated antibodies (4440 was used at 1:1,000 dilution), normalized by protein content, overnight at 4°C. The immunoprecipitates were separated by 6.25% SDS-PAGE and immunoblotted with a different, affinity-purified rabbit KDM5B antibody.
