Anti-Tartrate Resistant Acid Phosphatase (TRAP/ACP5) Antibodies

This range of mouse monoclonal antibodies was generated against tartrate resistant acid phosphatase (TRAP) and recognizes epitopes throughout TRAP.

Tartrate-resistant acid phosphatase (TRAP) is a glycosylated monomeric metalloprotein enzyme commonly expressed in mammals, that can be used as a biomarker for multiple clinical and basic applications. It has been used as cytochemical marker to identify hairy cell leukemia (HCL) cells as well as a marker for differentiated cells of monocyte lineage such as macrophages, osteoclasts and dendritic cells. TRAP is also used as a serum biomarker for bone disease by measuring elevated levels released from osteoclasts during bone resorption. Other increases in TRAP expression can conclude Gaucher's disease, HIV-induced encephalopathy, and osteoporosis. TRAP consists of two isoforms (Type 5a and 5b) that have different biochemical, clinical and biological properties.

mAb Recognition Western Blots and Immunoprecipitates

A: Western blots of dendritic cell lysates labeled with anti-TRAP mABs showing isoform specificity mAb 220 and 89 bound specifically to isoform 5a, where as 95 bound to both isoforms (35kDa 5a band and 16 kDa 5b band). MAb 15G6, 162 and 203 were unreactive. The uniform high-MW bands are due to non-specific streptavidin - biotin interactions and serve as a loading control. B: Immunoprecipitates of dendritic cell lysates using anti-TRACP mABs subjected to western blot and detected with mAb9C5. MAb14G6, 203, 163 under native conditions, precipated both isoforms. 220 and 89 under native conditions precipitated only isoform 5a. 9C5 reacts with denatured epitope of TRAP, showing reactive with TRAP5a-like precursor.
